That security is there to protect brain tissue from dangerous pathogens and foreign molecules, but it's also a barrier to the entry of therapeutic agents. However, there could be a way to sneak drugs past those barriers by opening them transiently, explains Dr Matthew Campbell from Trinity College Dublin. He's part of a team pioneering an approach that uses specifically designed small interfering RNA to suppress production of a protein called claudin-5, a key component of the barrier that stops drugs moving across from the bloodstream. Injecting siRNA systemically has been shown to improve the permeability of blood cells around brain tumours called gliobastomas in pre-clinical models. "Given the life-threatening nature of this sort of condition, systemic administration of siRNA targeting claudin-5 to allow periodic drug access to tumours would be ethically justifiable," says Dr Campbell. The approach could also be used in the treatment of traumatic brain injury, where suppressing claudin with siRNA could open the blood-brain barrier and encourage the movement of water away from the injury site, thereby reducing the potentially lethal effects of swelling, he added.   • SENS5 - Lysosomal enhancement strategies for the treatment of oxysterol-induced cytotoxicity © SENS Foundation 2011 - The Fifth SENS conference - Jacques Mathieu Authors JM Mathieu, PJ Alvarez Rice University 7-ketocholesterol (7KC) is a cytotoxic oxysterol that plays a role in many age-related degenerative diseases. 7KC formation and accumulation occur in the lysosomes in a number of cell types, hindering enzymatic transformation, and increasing the chance for lysosomal membrane permeabilization. We assayed the potential to mitigate 7KC cytotoxicity and enhance cell viability by transiently transfecting human fibroblasts to overexpress several 7KC-active enzymes. One of our engineered constructs, a lysosomally-targeted cholesterol oxidase that lacked isomerization activity, significantly increased cell viability at concentrations up to 50 µM 7KC relative to controls. Additionally, both overexpression of the lysosomal membrane protein LAMP1 and treatment with 0.9% hydroxypropyl β-cyclodextrin attenuated 7KC-mediated cell death. The overexpression of cholesterol oxidase, sterol 27-hydroxylase, and 11β-hydroxysteroid dehydrogenase, which were localized to the cytoplasm, mitochondria, and endoplasmic reticulum respectively, resulted in either decreased cell viability or no significant change from controls. These results indicate that the lysosome should be the target for treatment of oxysterol-mediated toxicity, and support the utility of using novel lysosomally-targeted enzymes for therapeutic benefit.

  • How many birds does it take to change the SWR? How many birds does it take to change the SWR? This is just a bit of fun looking at the very small but consistent changes that a group of swallows make to the Voltage Standing Wave Ratio (SWR) on my home made 20 metre antenna. It's not a scientific experiment but the results were, not surprisingly, reasonably consistent as it's a well know fact that you can use capacitive loading to alter the characteristics of an antenna. I did toy with the idea of arranging bird food at the ends of the elements were their capacitive effect would be more noticeable but I didn't want to encourage the crows as if they decide to land on the beam en masse as I think they would do more damage than just transiently shift the SWR! Right now, I just want to quietly sleep in a translucent wind embraced in wings. I'm looking for the whereabouts of the destiny I was entrusted with. A swarm of falling petal-like feathers dance down. Someday, surely, I'll reach the place that will be able to heal my lonesome heart. My eyes reflect that place before me as I stare upwards into the sky. I've gathered the fragments of my sadness. Eternity is so endless and suffocating. So much that it sounds like something someone would long for. My dream is but a mirage. But now, somewhere it's secretly and transiently blooming.
