All DNA polymerases require a primer to start DNA synthesis. The primer is formed inside of cells by a special RNA polymerase known as primase. (RNA polymerase does not require a primer) 3. DNA replication proceeds by two distinct mechanisms (both 5'-3', however)- one on each strand. Leading strand and lagging strand synthesis occur by different mechanisms, but both are catalyzed by the same DNA replication complex (Pol III, in the case of E. coli). 4. Leading strand synthesis is continuous in the 5' to 3' direction. Lagging strand synthesis can only occur when the leading strand synthesis opens up a new single stranded region for replication.   • Topoisomerase (topo I, top I) presentation in HD DNA topoisomerases are enzymes that introduce "swivel points" in the DNA helix to change its tertiary structure. Topoisomerases break one or both DNA strands, pass the strand(s) through the break, and rejoin them without using ATP. Topoisomerase I (Topo I) cuts a single strand of the double-helix. The unbroken (intact) strand then passes through the break. Finally, the ends of the initially cut strand are ligated. Eukaryotic topo I and topo II can remove both negative and positive supercoils. DNA can be relaxed (B-form double-helix) or supercoiled (when the double-helix coils upon itself). Anti-cancer agents camptothecin and etoposide, that respectievly target topo I and topo II, are used in chemotherapy. By inhibiting specific steps in the action of topoisomerases, the drugs convert them into DNA-breaking agents and lead to cell death. Human topoisomerase I is the molecular target of a diverse set of anticancer compounds, including the camptothecins. These compounds bind to transient topo I-DNA covalent complex and inhibit the resealing of a single-strand nick that the enzyme created to relieve superhelical tension in duplex DNA.   • Fluoroquinolones: Mechanisms of Action and Resistance Developed and produced by Animation Description In this animation, we demonstrate the biology of DNA replication leading to bacterial cell division in a gram positive bacterium, such as S. pneumoniae. The DNA is shown as a circular double strand within the bacterial cell. Like the DNA of all living organisms, it contains the unique genetic code for all of the proteins required for bacterial survival. Bacteria replicate by a process known as binary fission whereby one bacterium separates into 2 new daughter cells. However, before this can occur, the bacterium must make an identical copy of its complete circular DNA. DNA replication requires that the two strands of DNA separate so that the genetic code of the bacterium can be read and a new complimentary strand can be created for each of the original strands. To accomplish this, various enzymes known as helicases break the hydrogen bonds between the bases in the two DNA strands, unwind the strands from each other, and stabilize the exposed single strands, preventing them from joining back together. The points at which the two strands of DNA separate to allow replication of DNA are known as replication forks. The enzymes DNA polymerase then move along each strand of DNA, behind each replication fork synthesizing new DNA strands (in red) complementary to the original ones.
