The molecular tool used to cut DNA is a restriction enzyme such as EcoRI. The enzyme has a precise shape that allows it to run along the groove of the double helix, scanning for the base letter sequence GAATTC EcoRI then cuts the plasmid at this specific point... ...allowing a new piece of DNA to be inserted. When it cuts, EcoRI leaves a sticky end, which helps the new gene to attach. The joins are THEN stitched together by another enzyme called DNA ligase. The genetically engineered bacteria is then grown in a culture medium. Very quickly, large numbers of the bacteria can be produced, each with a copy of the inserted gene. The bacteria duly manufacture whatever protein the gene codes for, and so the desired product is produced.

  • Karen Hunter on Recombinant prion protein induces a new transmissible prion disease... Prion disease is a neurodegenerative malady, which is believed to be transmitted via a prion protein in its abnormal conformation (PrPSc). Previous studies have failed to demonstrate that prion disease could be induced in wild-type animals using recombinant prion protein (rPrP) produced in Escherichia coli. Here, we report that prion infectivity was generated in Syrian hamsters after inoculating full-length rPrP that had been converted into the cross-β-sheet amyloid form and subjected to annealing. Serial transmission gave rise to a disease phenotype with highly unique clinical and neuropathological features. Among them were the deposition of large PrPSc plaques in subpial and subependymal areas in brain and spinal cord, very minor lesioning of the hippocampus and cerebellum, and a very slow progression of disease after onset of clinical signs despite the accumulation of large amounts of PrPSc in the brain. The length of the clinical duration is more typical of human and large animal prion diseases, than those of rodents. Our studies establish that transmissible prion disease can be induced in wild-type animals by inoculation of rPrP and introduce a valuable new model of prion diseases.

English: In 1973, two scientists named Boyer and Cohen developed a way to take DNA from one organism and put it in the DNA of bacterium. This process is called recombinant DNA technology. First, a circular piece of DNA called a plasmid is removed from a bacterial cell. Special proteins are used to cut the plasmid ring open. With the plasmid ring open, the gene for insulin is inserted into the plasmid ring and the ring is closed. The human insulin gene is now recombined with the bacterial DNA plasmid. Many plasmids with the insulin gene are inserted into many bacterial cells. The cells need nutrients in order to grow, divide, and live. While they live, the bacterial cell processes turn on the gene for human insulin and the insulin is produced in the cell. When the bacterial cells reproduce by dividing, the human insulin gene is also reproduced in the newly created cells. Human insulin protein molecules produced by bacteria are gathered and purified.

Although FDA has approved two rhBMP products for very specific medical conditions, the safety and effectiveness of rhBMP in the cervical spine have not been demonstrated, and FDA has not approved it for this use. Over the past four years, FDA has received about 40 reports of complications associated with using rhBMP in the cervical spine. They involved swelling of neck and throat tissue, which resulted in compression of the airway or neurological structures in the neck. Some reports describe difficulty swallowing, breathing or speaking. Most of the complications occurred between 2 and 14 days following surgery. When airway complications occurred, emergency medical intervention was frequently necessary. FDA recommends that practitioners either use approved alternative treatments for cervical spine fusion, or consider enrolling as investigators in approved clinical studies. Patients treated with rhBMP in the cervical spine should seek medical attention immediately if they have symptoms of an airway complication, such as difficulty breathing or swallowing, or swelling of the neck, tongue, mouth, throat, shoulders or upper chest.   • Insidermedicine In Depth - November 4, 2010 - Recombinant Activated Factor VII Using an enzyme known as recombinant activated factor VII to stem life-threatening bleeding can place patients at increased risk for thromboembolic events, according to research published in the New England Journal of Medicine. Here is some information about a thromboembolic event: • It is the development of a blood clot, or thrombus, that develop in a blood vessel, either an artery or vein • The blood clot may then dislodge and begin to travel through the circulatory system. This is known as an embolism • The blood clot may then lodge in a major organ, including the heart, lungs, or brain, causing severe damage and even death. Researchers from the University of Amsterdam ***yzed data taken from 35 controlled trials that explored the risk of thromboembolic events among individuals given VIIa. These studies included over 4000 individuals who were given the drug to treat uncontrolled bleeding and nearly 350 healthy volunteers who took the drug to test its effects. Overall, 11% of the participants developed a thromboembolic event. Thromboembolic events occurring in the arteries occurred more frequently among those who received VIIa than those who received a placebo. This was particularly notable among elderly individuals. There was no difference in the rate of thromboembolic events occurring in the veins between those who received VIIa and those who received a placebo. The in vitro expression and electrophysiological recording of recombinant voltage-gated ion channels in cultured human embryonic kidney cells (HEK-293T) is a ubiquitous research strategy. HEK-293T cells must be plated onto glass coverslips at low enough density so that they are not in contact with each other in order to allow for electrophysiological recording without confounding effects due to contact with adjacent cells. Transfected channels must also express with high efficiency at the plasma membrane for whole-cell patch clamp recording of detectable currents above noise levels. Heterologous ion channels often require long incubation periods at 28°C after transfection in order to achieve adequate membrane expression, but there are increasing losses of cell-coverslip adhesion and membrane stability at this temperature. Mocarski, Jr., of Stanford University Medical School, builds upon his lecture in Part One of the series (Core Concepts of Microbiology) by explaining the basic recombinant DNA and gene-cloning methods used in the field of biotechnology. Mocarski explains how the universality of the genetic code makes it possible for scientists to recombine DNA, that is, take DNA from one organism and move it into another. He also explains how recombinant DNA concepts are used in the "expression" of human proteins into bacteria, a process in which a human coding sequence is taken and inserted into a bacterial context, allowing the bacteria to produce abundant supplies of a "foreign protein" (foreign to the bacteria) that can be of commercial and therapeutic use.
