  • Compounds for PhotoDynamic Therapy FINAL.mpg Porphyrinoids are used as photosensitizers for photodynamic therapeutic (PDT) studies. A goal is to develop high-yield reactions to append porphyrinoids with 4-8 copies of an array of biomolecules or biocompatible molecules. For example, four non-hydolysable glucose moieties on this porphyrin drives the selectivity to several breast cancer cell lines such as MDA-MB-231, and low power visible light causes both cell necrosis and apoptosis. Innovation. This work aims to developed make conjugates to porphyrinoids missing one double bond on a pyrrole (chlorin Chl), or two double bonds on adjacent pyrroles (isobacteriochlorin Ibc) or opposite pyrroles (bacteriochlorin Bc). Phthalocyanines (Pc) will be used as well. The same substitution chemistry on both of these systems should yield an array of conjugates. These conjugates will be designed to have three functionalities: (a) the R-groups impart high selectivity and greater uptake to cancer cells over normal cells, eg sugars, peptides, antibodies; (b) the photophysical properties are tunable -- for maxima absorption and formation of the triplet state for singlet oxygen sensitization; (c) the metal ion can be a radionuclide for imaging and/or therapeutics.
