- Passaging Human Pluripotent Stem Cells Using STEMium® and StemDS® SCRIPT: In this video, we will demonstrate how to use STEMium- our proprietary serum-free, feeder-free human pluripotent stem cell growth medium and StemDS- a specially formulated stem cell dissociation solution. The following procedure was developed for passaging human pluripotent stem cells, splitting in a 1 to 3 ratio. Please keep in mind that further optimization may be needed for other cell lines. It is also important that users become thoroughly familiar with the entire method before proceeding, as culturing human pluripotent stem cells may require different laboratory techniques than other media and cell lines. List of reagents : The following reagents are required: BD Matrigel coated plate, instructions for this preparation can be found in our online video library, DPBS, complete STEMium, StemDS, pipette tips, serological pipettes, pipettors, laminar flow hood, 37 degree celsius 5% carbon dioxide incubator and an inverted microscope. Microscope zoomed up video and 2 still images of healthy cell: Prior to passaging, cells should be examined daily under an inverted microscope for overall confluency and differentiation. (image 1 and 2) Morphology of H9 human embryonic stem cells grown under feeder-free condition and in STEMium will be different compared to other media. image 3 Confluent still image Cells should be subcultured when they remain in log phase and cell colonies are nearly touching, between 70 to 75 percent confluent, as suboptimal passaging may result in ...

In this study, the researchers reprogrammed ordinary fibroblasts, cells that contribute to scars such as those resulting from a heart attack, converting them into stem cells that fix heart damage caused by infarction. The findings appear in the current online issue of the journal Circulation. Timothy Nelson, MD, Ph.D., first author on the Mayo Clinic study, talks about the study and it's findings.

- Dr. Uma Lakshimipathy presents Generation of Transgene-Free Induced Pluripotent Stem Cells Learn more at Dr. Uma Lakshmipathy presents work on the creation of integration-free induced pluripotent stem cells at high efficiency with Sendai Virus using the CytoTune™ -iPS Sendai Reprogramming Kit. [audio transcript] So to start with the most common method for generating iPSC is transduction of the four factors and shown here is the Yamanaka factors and after a black box in which takes anywhere between three to four weeks you end up with iPSC colonies. So the biggest bottleneck right now; one is the efficiency of iPSC formation depending on what kinds of cells you start with, the efficiency is really low. And the second thing, the second bottleneck is how do you detect these emerging iPSC colonies depending on the expertise of the users and if you look for pluripotent stem cells, people can either pick it really easily or there is always like an issue on what clones you place your bet on. So when it comes to efficient methods, there are several methods starting from viral non-integrating and more small molecule methods such as mRNA, microRNA and small molecules. But one of the methods we really worked on is a non-intergrating viral method based on Sendai virus. The reason why this method is superior to the other current methods is its efficiency; it's based on Sendai virus which is a RNA virus. So again coming back to the different methods that are out there, if you were to... this is a graph I took from a published paper... if you look at the ...

- Induction of Pluripotency by Defined Factors Air date: Thursday, January 14, 2010, 3:00:00 PM Timedisplayed is Eastern Time, Washington DC Local Category: Wednesday Afternoon Lectures Description: Human ES cells have been expected as suitable resources for cell transplantation therapies. However, it has sparked ethical controversy and causes immune rejection. Hence, we decided to generate an ideal pluripotent stem cell for innovative medicine. At first, we constructed a pluripotency assay system that the candidate factors are introduced into neonate fibroblasts via retrovirus vectors. As the result, the set of Oct3/4, Sox2, c-Myc, and Klf-4 gave rise to drug resistant colonies implying potential pluripotency. The survived cells resembled ES cells in terms of morphology and proliferation showed ES cell markers and formed teratoma. It was named as induced pluripotent stem cell (iPS cell). iPS cells were created even from adult mouse fibroblasts. Moreover, iPS cells based on Nanog-expression demonstrated germline transmission. Furthermore, we successfully generated iPS cells from human adult fibroblasts, using a modified protocol. However, tumor formation was observed in chimera mouse, probably due to c-Myc retrovirus integrated into genome. We re-modified the protocol and successfully established iPS cells without using c-Myc. As further effort to lower a risk of tumorigenesis, we recently succeeded in developing a virus-free method - using a pair of plasmid vectors, instead of retrovirus vectors, to introduce the ...
