  • D3bx An 8-cell human embryo with average morphology, biopsied roughly 70 hours (Day 3) post-fertilization. When indicated, a single cell (blastomere) can be removed from a Day 3 embryo and subsequently tested for inheritable genetic diseases. The embryo is returned to culture and the biopsied cell is couriered to a genetic lab. Results from the biopsied cell are generally available inside of 48 hours. The embryo is in a solution of calcium and magnesium-free solution that is necessary to loosen the cell-cell junctions and isolate a single nucleated cell (no nucleus, no DNA, no diagnosis). The first pipet applies an acidified solution to the protein coat (zona pellucida) that dissolves a hole approximately 2/3 the diameter of the blastomere. The embryo is then moved away from the locally acidified area. It is also possible to use a laser to 'hatch' a slit through the zp, but not every IVF lab has access to one. The second tool is pushing a minute amount of culture media through the space in the zp to create just enough positive pressure to extrude a single cell. Using a positive-pressure biopsy technique has the advantages of being potentially very quick, unintrusive and with less likelihood of shearing cells and subsequently carrying-over potential DNA contamination between successive embryos. It has also been especially helpful on microscope-rigs with less-than-perfect optics or with poor vibration isolation.

Those who knew Ben, knew that he never shied away from trying something new, not even video games. One day his mother took him to see a doctor, and while he was waiting, he played with his Game Boy. When the doctor walked in and saw Ben, he looked at his chart and walked back over to look at his eyes. The doctor looked down to see if he was really playing the game and exclaimed to his mother, "His eyes are nucleated." Ben's mother replied, "Oh yes, he's blind."   • Giant Planet Formation - Jack Lissauer (SETI Talks) SETI Talks Archive: The observed properties of giant planets, models of their evolution, and observations of protoplanetary disks all provide constraints on the formation of gas giant planets. The four largest planets in our Solar System contain considerable quantities of hydrogen and helium, which could not have condensed into solid planetesimals within the protoplanetary disk. The preponderance of evidence supports the core nucleated gas accretion model of formation of the giant planets. According to this model, giant planets begin their growth by the accumulation of small solid bodies, as do terrestrial planets. However, unlike terrestrial planets, the growing giant planet cores become massive enough that they are able to accumulate substantial amounts of gas before the protoplanetary disk dissipates. During this talk, Dr. Lissauer will present the first models of giant planet formation that account for both the planet's internal energy budget and gas flows within the protoplanetary disk.
