Part 2: SOX2 ***ysis in NSCLC In this talk, a study on SOX2 amplification in non-small cell lung cancer with a special focus on IHC assessment using the Definiens Tissue Studio image ***ysis software will be presented. The aim of the study was to verify SOX2 amplification and protein over-expression in non-small cell lung cancers (NSCLC) and to compare these results with patient and tumor characteristics. A total of 940 NSCLCs from two independent population-based cohorts containing predominantly adenocarcinomas of the lung and squamous cell lung carcinomas, were assessed by fluorescence in-situ hybridization (FISH) and immunohistochemistry (IHC) to study SOX2 amplification and expression level. For protein expression ***ysis, the expression level was explored by utilizing Definiens Tissue Studio.

Mycosis fungoides and the Sezary syndrome are cutaneous T-cell lymphomas. These are characterized by the expansion of a clone of malignant CD4 helper T-cells that have a propensity to home and accumulate in the skin, lymph nodes, and peripheral blood. Confirming the diagnosis of mycosis fungoides is difficult in the early stages of the disease. The clinician will biopsy patch stage lesions and ask the pathologist to rule out MF. The classic features of the disease are usually absent at this stage. Typically a few atypical lymphocytes are seen and the pathologist reports an atypical lymphocytic infiltrate, suspicious for MF. Biopsy in patch-or-plaque stage shows a lymphocytic infiltrate in the superficial dermis, with individual lymphocytes migrating among epidermal keratinocytes. The affinity of the malignant T-cells to migrate to the epidermis is called epidermotropism. The lymphocytes are pleomorphic, hyperchromatic, and have hyperconvoluted nuclei. Staining of skin-biopsy specimens with a panel of lymphocyte markers is used sub classify the clone of cells. Immunohistochemistry and molecular genetics have demonstrated aberrant expression of the T lymphocyte surface antigens. fly93to09 Dissection of the Adult Drosophila CNS. This is a start-to-finish video showing the dissection of a fruit fly brain. The video begins by introducing the tools and materials and then goes through the process step by step. The method is suitable for downstream staining (eg immunohistochemistry or enzymatic) and is intended to be quick. Because the dissection is so quick, it can be (and is) performed in paraformaldehyde fixative - fixing two birds with one stone as it were. There is always a risk of distortion to the brain during dissection (by stretching) which is somewhat minimised by fixation. Most of the materials should be available in a basic laboratory - the most critical is very very sharp forceps (tweezers). The video shows a sharpening stone and you should sharpen your own tools for the best results. The video has been converted from an old 1993 VHS format with no soundtrack but a written protocol and associated example images have been on-line since the 90's at which I believe makes it one of the earliest on-line protocols in bioscience. Comments welcomed even though I have moved up the evolutionary scale to work on arabidopsis now :) . .

Immunihistochemistry on Paraffin Sections of Mouse Epidermis Using Fluoresent Antibodies part 1 Abstract: In the epidermis, immunohistochemistry is an efficient means of localizing specific proteins to their relative expression compartment; namely the basal, suprabasal, and stratum corneum layers. The precise localization within the epidermis of a particular protein lends clues toward its functional role within the epidermis. In this chapter, we describe a reliable method for immunolocalization within the epidermis modified for both frozen and paraffin sections that we use very routinely in our laboratory. Paraffin sections generally provide much better morphology, hence, superior results and photographs; however, not all antibodies will work with the harsh fixation and treatment involved in their processing. Therefore, the protocol for frozen sectioning is also included. Within paraffin sectioning, two fixation protocols are described (Bouin s and paraformaldehyde); the choice of fixative will be directly related to the antibody specifications and may require another fixing method.

Immunihistochemistry on Paraffin Sections of Mouse Epidermis Using Fluoresent Antibodies part 2 Abstract: In the epidermis, immunohistochemistry is an efficient means of localizing specific proteins to their relative expression compartment; namely the basal, suprabasal, and stratum corneum layers. The precise localization within the epidermis of a particular protein lends clues toward its functional role within the epidermis. In this chapter, we describe a reliable method for immunolocalization within the epidermis modified for both frozen and paraffin sections that we use very routinely in our laboratory. Paraffin sections generally provide much better morphology, hence, superior results and photographs; however, not all antibodies will work with the harsh fixation and treatment involved in their processing. Therefore, the protocol for frozen sectioning is also included. Within paraffin sectioning, two fixation protocols are described (Bouin s and paraformaldehyde); the choice of fixative will be directly related to the antibody specifications and may require another fixing method.
