  • AutoFill with Cell Paper recipe applied to Synaptic Cleft tomogram v2: Cytoplasm and AO Alas, the rendering with Ambient Occlusion and Cytoplasm. This is an awful recipe attempt for Cytoplasm, but I'm building a new version filled with lots of actin filaments to better reflect the cytoplasm of the presynaptic cell. This version shows a more accurate recipe for the vesicle membranes. Although the geometry is still just raw from AutoFill, I took a few seconds to light it better for clarity. Here you can see all of the dominant structures pulled down from the Cell Paper mentioned below. See how crowded and interdigitating the vesicles are when fully decorated with proteins. This version of Autofill has an issue where vesicles can't see neighboring vesicles unless they already have proteins on their surfaces, so some of the first proteins places, (ATP synthases, end up poking through their neighbors.) The algorithm solves that problem, but in this lazy setup, it doesn't see the vesicles as separate polyhedrons- must implement that in the next version... Next I'll post the same model with the cytoplasm turned on. I'm working on another model with weekend that has actin dominating the cytoplasm instead of all the placeholder GroELGroES complexes.   • Plant Cell Motility and Laser Microsurgery of Cytoplasmic Strands Plant cells are shaped by rigid cell walls. Osmotic forces press the plasma membrane tightly against these walls. The walls can be removed with enzymes. The remaining structures, the protoplasts, are now bordered by the plasma membrane. Since the shaping force of the wall is missing, isolated plant protoplasts are usually perfectly spherical in shape as a result of non-directional osmotic forces. However, not all isolated plant protoplasts are spherical. Through cytoplasmic streaming, cytoplasmic strands continuously change their length, branching and attachment, and as a result the shape of the protoplast may change. A laser microsurgery experiment provides proof that the inner tension built by the cytoplasmic network is moving the cell. A laser beam focused at a cytoplasmic strand will cut the strand and cause the network to collapse without destroying the integrity of the cell. As a result osmotic forces can take over and round the protoplast. Moving around is not an option for most cells of higher plants, and cytoplasmic streaming serves normally the intracellular transport - but they have not totally lost the basic skills that would enable them to move (entered in www.Chlorofilms.org video contest).

  • Cytoplasmic streaming in Elodea Cytoplasmic streaming is the flowing of cytoplasm in eukaryotic cells. This occurs in both plant and animal cells. It creates cytoplasmic reorganization during cell reproduction. In some unicellular eukaryotes, such as amoeba, it provides the mechanism for cell locomotion. It aids in the delivery of nutrients, metabolites, and genetic information to all parts of larger plant cells. It vigorously "stirs" these components about the cytoplasm, allowing them to flow to all parts of the cell. As membrane-bound organelles crawl along the inner cell wall they set the cytoplasm into motion. This streaming allows other organelles and nutrients to flow in the stream created. Actin filaments play a role in streaming. Cyclosis is the circulation or streaming of the cytoplasm within some living cells. In plant cells, chloroplasts may be moved around with the stream. The rate of motion is usually affected by light exposure, temperature, and pH levels. Chloroplasts make use of cytoplasmic streaming to move to optimum position within the cell for maximum light absorption used in photosynthesis. This is presumably what is pictured in the video.
