Mary Beckerle (Univ Utah) Part 2: Discovery and Characterization In the second segment, I describe the identification of the focal adhesion protein, zyxin, by my lab. Recent work revealed that zyxin is down-regulated upon expression of the Ewing sarcoma oncoprotein, EWS-FLI. Loss of zyxin expression results in enhanced cell motility and is also associated with failed apoptotic signaling. Evidence that zyxin shuttles between focal adhesions and the nucleus is presented. The impact of reduced zyxin expression on tumor progression is discussed. See more at

SFCM 10/11 12: Mechanical characterization of wafer Silicio Multicristalino para Aplicaciones Fotovoltaicas Mechanical characterization of wafer equivalents from wire-sawing to thin film deposition TERESA ORELLANA Fraunhofer Institute for Solar Energy Systems (ISE) Freiburg, Alemania The epitaxial wafer equivalent concept at Fraunhofer ISE promises product cost reduction by depositing 20 μm crystalline silicon thin film by CVD on a low cost silicon substrate [2].The substrate of the wafer equivalent should provide a good mechanical strength while the epitaxial layer is responsible for the electrical performance of the final solar cell (Epi-Cell). At Fraunhofer ISE, multicrystalline silicon blocks between 20 kg and 250 kg are crystallized via directional solidification. The crystallization is done in a Vertical Gradient Freeze (VGF) furnace with graphite heaters. Silicon is put into a fused silica crucible (SiO2), which is coated with a silicon nitride (Si3N4) liner to prevent a chemical reaction between crucible and silicon. Multicrystalline high boron doped silicon and up-graded metallurgical silicon (UMG-Si) are crystallized for manufacturing the substrate of the epitaxial wafer equivalent, thus saving the costs for high purity silicon material. Using silicon as a substrate reduces problems arising from differences in thermal expansion coefficients and misfit of lattice parameters. The Epi-Cell can also benefit from manufacturing cost reduction if the wafer equivalent presents a sufficient mechanical strength to ensure ...

Time-Lapse Characterization of Arabidopsis Starch Metabolism Mutants During the early exponential growth phase of plants, the majority of photoassimilates are reinvested into new leaf material. Leaf area, one of many growth parameters, has been examined by time-lapse photography in Arabidopsis starch metabolism mutants. The irregular shape of leaves can make measurement by traditional means difficult, especially for large numbers of plants. By taking a digital picture of the leaves to be measured, along with an object of known size, the pixel area of the leaves can be can quantified into physical units. This is accomplished using the color range and histogram tools in Adobe Photoshop. A screen capture demonstration of this process is available on YouTube ( The accompanying song, Claire de Lune, was composed by Claude Debussy and performed by pianist Van Cliburn. Protein Characterization II This course is part of a series taught by Kevin Ahern at Oregon State University on General Biochemistry. For more information about online courses go to ecampus.oregonstate.edu 1. Nucleic acids (which are negatively charged and 'rod-like' in shape) can be separated by agarose gel electrophoresis readily. In this technique, a 'gel' is made that consists of a matrix material (agarose) that forms a sort of 'mesh' of hole through which the DNA molecules pass. Electric fields are used to separate macromolecules by size. The sample is loaded on the top of the gel and electrical current is passed through it such that the bottom electrode is positive and the top one is negative. Negatively charged DNA molecules at the top of the gel are driven away from the top towards the bottom. Small molecules make their way through the gel fastest and big molecules travel more slowly. The key to gel electrophoresis is to have all of the molecules being separated have a negative charge. 2. Proteins, however are usually globular in their native state and, without other modification, may be negatively, positively OR neutrally charged. Another type of electrophoresis employs polymers of acrylamide (called polyacrylamide) to form the mesh of the gel. This mesh has smaller holes that agarose and allows separation of smaller molecules like proteins. To separate proteins on the basis of size, the detergent sodium dodecyl sulfate (SDS) is add to the protein mixture, causing the proteins to denature ...

Protein Characterization I This course is part of a series taught by Kevin Ahern at Oregon State University on General Biochemistry. For more information about online courses go to ecampus.oregonstate.edu 1. Folding of proteins is dictated by the sequence of amino acids. Folding does not occur randomly. If it did, proteins would take longer than the age of the universe to fold and instead they fold in seconds. This is known as Levinthal's paradox. 2. The tendency of each amino acid to participate in alpha helices, beta strands/sheets, and turns are known and this information can be used to predict secondary structure of a protein from the amino acid sequence. 3. Misfolding of proteins can have disastrous consequences (but not necessarily always). One disastrous consequence is that of prions, which are "infectious" proteins implicated in diseases, such as mad cow disease and Creutzfeld-Jacob disease in humans. Each of these is a brain-wasting disease the results from misfolding of a brain protein known as PrP. The misfolded protein apparently helps convert properly folded proteins to the misfolded state and cause the disease. 4. Cells use chaperonin complexes to help insure proteins fold properly. Chaperonins are induced by heat shock of cells. Protein Purification 1. Purification of proteins exploits differences in charge, size, shape, and affinity for specific compounds. Centrifugation (artificial gravity) provides a means of precipitating cellular components. The faster one spins the rotor of the ...
