They bind to and inactivate foreign molecules—the antigens—and mark them for destruction. Each IgG molecule consists of two light chains and two heavy chains. The heavy chains have carbohydrates attached. The regions of the antibody that bind to antigens are located at the very tips of the two arms. Each arm of the antibody is composed of four domains. Two are called the variable domains, contributed by the heavy and light chains, and hence called VH and VL. The variable domains are attached to two constant domains, again one each from the heavy and light chains, and hence called CH and CL. Variable and constant domains share a similar structure, called the Ig fold. Each domain consists of a pair of beta sheets, one with three strands and one with five. A single covalent disulfide bridge holds the two sheets together, which results in a rigid and very stable domain. Specifically, this is a rotating Immunoglobulin G, IgG (PDB ID 1IGT), which is the dominant class of antibodies found within the body and also the first to be structurally ***yzed through X-Ray Crystallography. The holes found within the upper Y branches are the sites where the antibody binds the antigen substrate and are known as the Fab regions for antigen-binding fragments. The lower Y branch is called the Fc region for crystallizes readily. These holes have a 3-d structure, charge density, ability to H-bond, and hydrophobic regions that is created by the unique pairing of a heavy chain (blue) with a light chain (yellow), which is determined by the amino acid residues found in that particular antibody. The heavy and light chains are linked by covalent and disulfide bonds. These pockets can only recognize a particular molecular structure found on the antigen. It is this high degree of a lock and key fit that enables the great specificity by which antibodies bind a type of antigen. The instrument uses surface plasmon resonance, a phenomenon in which the intensify of reflected light attenuates as the laser incidence angle changes, to measure antigen-antibody reactions simply. Antibodies are affixed to the tip of a glass sensor probe, which is 1.5mm in diameter and 50mm in length, and the probe tip is submerged in a measurement sample. The sample can be as little as 10 microliters. The instrument is 20cm long and 10cm wide, small enough to be carried around. The research group plans to make the instrument smaller and even more accurate and develop more applications for commercializing it.

X-ray crystallography is a method of determining the arrangement of atoms within a crystal, in which a beam of X-rays strikes a crystal and scatters into many different directions. From the angles and intensities of these scattered beams, a crystallographer can produce a three-dimensional picture of the density of electrons within the crystal. From this electron density, the mean positions of the atoms in the crystal can be determined, as well as their chemical bonds, their disorder and sundry other information.   • ASCO GI: Antibody Slows Metastatic Colon Cancer ORLANDO -- Patients with nonmutated colorectal tumors had significant improvement in progression-free survival (PFS) when the monoclonal antibody panitumumab (Vectibix) was added to conventional chemotherapy, data from two randomized clinical trials showed. When used in first-line therapy for metastatic cancer, the antibody-chemotherapy combination was associated with a 20% improvement in the hazard ratio for progression compared with chemotherapy alone. In the second-line metastatic setting, the combination improved the hazard ratio by 27%.
